Compact and efficient bench-top Lyophilizer / Freeze Dryer should have following specifications and features:

8. Laboratory Water Purification System
Complete system including analytical grade water unit with storage tank and ultra pure water unit with dispenser.

The system should be capable of handling feed water conductivity upto 2000 microsiemens/cm.

The system should comprise of a prefiltration unit and Ion exchange resin based units to produce Type-II quality water (corresponding to analytical grade water) as defined by CAP, NCCLS and ISO 3696 (BS 3997).

It should have two stage prefiltration system with a inlet screen filter, 5 micron and 1 micron wrapped type depth filter and a DC pump operating at less than 50 db noise levels.

Should be easy to use with low running costs, give constant flow rate in all seasons, with inorganic, organic and microbiological purity.

It should not need a softener or softening ion-exchange resin cartridges

It should have mixed bed ion electro deionisation module (electrically operated) with auto regeneration.

It should have coaxial resistivity cell with a flow through design and a cell constant of 0.01 cm-l.

It should have continuous electrodeionization integrated into the system to eliminate the need for costly regeneration and/or resin exchange.

Should have very less energy requirement.

System to be based on advanced reverse osmosis (OS) technology.

It should have pretreatment cartridge with antiscaling compound and silver impregnated carbon; pretreatment cartridge with 3 micron and 1-micron PP depth filter; High flux thin film composite polyamide RP membrane with 94-99% rejection; conductivity meter before and after RO to measure the performance of the system; recirculation loop with capillary tube and diaphragm valve; mixed bed ion electro deionisation module with auto regeneration; carbon beads at cathode of the EDI module to prevent scaling of the module; Premeate divert valve which will divert low quality water to the drain; pump with unique temperature feed back mechanism. Should have complete 3 step purification process using complementary technologies.

In the first purification step, the Pretreatment pack should remove particles (1 ?m filter), free chlorine and colloids from tap water (activated carbon filter), should include anti-scaling compound which prevents the reverse osmosis membrane from scaling from hard water.

In the second purification step, the Reverse Osmosis Unit should remove 95-99% of inorganic ions and 99% of all dissolved organic substances (MW > 100 Dalton) as well as microorganisms and particles. In the third purification step, Ion-exchange resin module should remove the remaining ions.

It should have autodiagnostics and self-maintenance functions like, display of quality parameters, routine maintenance reminders, self-maintenance functions like automatic flush, automatic rinse, automatic cleaning cycle.

After each purification step, relevant parameter is to be checked in the system.

Replacement of the pretreatment pack should be based upon the actual amount of water pretreated.

The performance of the unit should be as follows:

Flow rate: 10 litres/hour.

Resistivity of product water: > 5 M ½. cm (typically 10 to 15 M ½. cm).

Conductivity of product water: < 0.2 µS/cm (typically 0.067 to 0.10 µS/cm).

TOC typically: < 30 ppb.

Silicate content: > 99.9% retention.

Water recovery: 24%.

The unit should have 30 litres polyethylene reservoir with Tank Vent filter.

The reservoir should have sensor rod float switch, programmed to have high and low level cut off based on water level in tank.

The polyethylene reservoir should be designed to maintain consistent purity of stored water and provide effective protection against air borne contaminants.

The reservoir should fill from the bottom.

It should have provision for upgradability: distribution pump, UV sanitization module and water sensor.

The tank vent filter should be made of soda lime, activated carbon and 0.65 micron hydrophobic membrane to trap contaminants present in atmospheric air.

(b) Ultra pure water unit
Ultra pure water unit should produce water of following specifications.

Resistivity: 18.2 MΩ. cm at 250C

TOC: < 5 ppb

Particulates (??0.22 ?m): < 1 particulates/ml

Bacteria: < 1 cfu/ml

Pyrogens (endotoxins): < 0.001 EU/ml

Should be a compact unit with convenient and flexible dispensing options for drawing precise volume.

Should have provision to use unique range of final purifiers.

Should come with multicolor graphic display screen showing the details of systems operation and performance and the product water.

Ultra pure water unit should consist of pretreatment pack, double wavelength UV lamp which ensures organic molecule oxidation and bacterial destruction and polishing cartridge that removes ionic and organic contaminants below trace levels.

It should use technology that enables full traceability.

The ultra pure water produced by the system should recirculate through a loop to the delivery unit where the final required step occurs.

The measurement of both organic and ionic quality of produced water should occur at the outlet of the system through appropriate calibrated meters viz., accurate total organic carbon (TOC) monitor and high sensitivity resistivity cell.

It should have ultrafilter for removing particulates and bacteria.

It should produce pyrogen and nuclease-free ultrapure water suitable for biochemical and molecular biology applications.

Should have enhanced flexibility with convenient and adaptable dispensing.

It should allow sensitive ionic detection with high precision resistivity meter.

It should have built in TOC monitor with a 0.5 ml Quartz cell and UV lamp which accurately measures TOC online from 1-999 ppb.

The design of the resistivity meter and TOC meter should follow the suitability test requirements as per USP30- NF 25.

Should have on site validation protocols with IQ, OQ, MP and PQ using calibration equipment and qualification workbook.

TOC measurements should perform automatically on a continuous basis during production and intermittently during periods of non-use.

9. UV-VIS NIR Spectrophotometer
SPECIFICATONs

10. Real time-PCR machine
System capable of doing genotyping, SNP analysis, gene expression and quantification assays with multitasking facility. System shall provide online cycle-by-cycle monitoring with continuous display of reading for fluorescence, temperature change and progression of amplification. System should have a minimum of 5 excitation filters (450,483,523, 558 and 615 nm) and six emission filters/channels with CCD camera for signal detection.

Detection dyes – SYBR green, Fluorescein, Fam, Hex, Vic, Joe, Tamara, Cy5, LC red, Rhodamine, Cyan 500, DABCYL and T red etc.. The system shall have built in facility to use both 96 well and 384 well micro plates. The typical run time should be less than 60 minutes for 45 cycles with a temperature range of 40-95. the ramping rates with heating at 6.30C/Sec and cooling at 3.50C for ultra rapid cycling. The excitation source may be of high intensity halogen or xenon. System software should have the provision to use fit point method or second derivative maximum for calculating the concentrations. System shall be supplied with licenced probe/primer design software. The analysis workstation should be a branded Pentium IV PC.
11.Minus 200C Freezer
Microprocessor based upright type, environmental friendly convenient storage drawers with locks. The capacity is 450 lit with cooling range between minus 20 and 300C. The system shall have temperature and power failure alarm system. Zero adjustment via control panel. Independent top/bottom controls to meet differing freezer needs simultaneously. The freezer should have two independent freezer cabinets in one body with separate defrost and operations.

12.Sequencing type gel electrophoresis system
Apparatus designed for vertical polyacrylamide gel separation of oligo and polynucleotides, nucleic acid sequencing, genotyping and polymorphic studies. Dimension of the apparatus shall be 42 X 22 X 44 cm(approximately), glass plate size approximately 33 X 39 X 4.75 mm, working buffer volume 1 lit and Gel size 31 X 38 cm for 0.4mm gel. 13.Dot Blot/Slot Blot apparatus
